Mechanism of Action

Oxaliplatin works by binding to the DNA in cancer cells and forming cross-links. These cross-links interfere with the normal function of DNA, particularly its ability to replicate and repair. As a result, cancer cells are unable to divide properly and eventually die.

Unlike older platinum drugs, oxaliplatin forms different types of DNA adducts that may be effective even in cancers that have become resistant to other treatments. It is considered more potent and has a broader spectrum of action when combined with fluorouracil-based therapy.

Uses

Oxaliptin is used mainly in the treatment of:

  • Colorectal cancer, in both early-stage (after surgery) and advanced metastatic stages

  • Often part of the FOLFOX regimen, which includes fluorouracil and leucovorin

  • It may also be used off-label in other gastrointestinal cancers when prescribed by an oncologist
