Proguheal contains Proguanil hydrochloride, an antimalarial medication used for both treatment and prevention of malaria, particularly in regions where malaria caused by Plasmodium falciparum is common. It is often prescribed alone or in combination with other antimalarials, such as atovaquone.

Mechanism of Action
Proguanil works by interfering with the folate metabolism of the malaria parasite. It is converted in the body to its active form, cycloguanil, which inhibits the enzyme dihydrofolate reductase (DHFR). This enzyme is essential for DNA synthesis and cell multiplication in the parasite. By blocking this enzyme, Proguanil stops the malaria parasite from multiplying inside red blood cells, helping the body clear the infection.

Uses
Proguheal is used for:

  • Prevention of malaria, especially in travelers to malaria-endemic areas

  • Treatment of uncomplicated malaria, often in combination with other drugs like atovaquone

  • Resistance management, particularly in areas where resistance to other antimalarials like chloroquine is common

    Adverse Effects
    Proguanil is generally well tolerated, but may cause:

    • Mild gastrointestinal symptoms, such as nausea, vomiting, or abdominal pain

    • Mouth ulcers or hair loss, especially with long-term use

    • Skin rash or itching in some individuals

    • Rarely, blood-related side effects like anemia or leukopenia, especially in patients with underlying kidney problems
